Ambika Rao made her debut with the Malayalam film Kisan released in 2006 as Ammalu was the beginning of her professional career and earlier she was assistant director for the Malayalam film Krishna Gopalakrishna in 2002.
Her last film was a Malayalam film named Paapam Cheyyathavar Kalleriyatte, it was released in 2020 as Philomina. Ambika liked watching movies since childhood. Rao used to go to the cinema hall with her father and siblings to watch movies. After getting divorced at the age of 37 she returned back to the movies.
Before working in films, she worked as a data entry operator. Before becoming an assistant director, she was asked to teach Malayalam to actors. She helped Usha Uthup in her Malayalam songs as well.
Ambika Rao directed the first short film titled ‘Smarana’ in year 2016. The film was shot in her flat because she had a lesser budget to direct the film. Rao was also well known by the name ‘The Coach’ on the movie sets.
