Madhu Mantena was married to Masaba Gupta but they got separated in 2020. He has a cousin named Ram Gopal Verma who is also involved in the filmmaking business. Before Masaba he was married to Nandana Sen. He attended a local school in Hyderabad.

In early 2008, Madhu Mantena co-produced Ghajini which was the highest-grossing Indian film of the same year. Since then Madhu has produced movies such as the trilingual sleeper-hit Rakht Charitra in 2010, and the political thriller Rann in 2010 which screened in Toronto at International Film Festival.
Madhu has co-founded Phantom movies with Anurag Kashyap, Vikas Bahl, and Vikramaditya Motwane, which went on to deliver works such as Lootera in 2013, Queen in 2014 starring Kangna Ranaut which won the National Film Award for Best Feature movie in Hindi for that year, Hasee Toh Phasee in 2014. He was also behind the making of the Neo-Noir films like Bombay Velvet 2015 and Ugly in 2015 which screened in the Directors’ Fortnight section at the 2013 Cannes Film Festival and the New York Indian Film Festival. His movie Masaan screened in the Un Certain Regard section at the 2015 Cannes Film Festival winning two awards and also winning a National Film Award.
Madhu Mantena started his career by making his own music label as a teenager which he then sold to the Supreme Recording Company. Madhu then went on to set up Adlabs’ International Operations under the aegis of Manmohan Shetty and head of Saregama Films. Madhu has built media entities with different various companies and the media value chain, each specializing in the fields of talent management, content development, also content production, content distribution and monetization.
He has also co-founded KWAN, which is a celebrity management company with Anirban Das Blah who is the erstwhile CEO of Globosport. In 2012 KWAN announced a joint venture along with Creative Artist Agency (CAA) to form a new entity, CAA KWAN. His talent management agency currently commands a variety of business interests and manages a repertoire of Indian talent as well as an international talent.
