She was born on December 10, 1942, in Perth, Scotland. Her father was a bus driver. However, nothing much is known about her mother. She also has a brother named Brian Souter.

She did her schooling at Caledonian Road Primary School and Perth High School. Later, she qualified as a nurse.

She first married Robin Nicol Gloag, with whom she has a son named Jonathan. However, they both divorced. Later, she married businessman David McCleary. She also adopted a Kenyan boy named Peter and a 6-year-old Kenyan grandson.

Dame Ann Gloag spent 20 years as a burn unit sister. She helped create Africa Mercy (Mercy Ships), a floating hospital and worked on Mercy Ships UK.

Stagecoach acquired further bus companies in Scotland, Newcastle, and London, with the addition of Manchester in 1993.
